I have worked as a postdoctoral associate with Dr Dhaval Shah at University at Buffalo NY USA and during this tenure, I have worked on developing novel protein therapeutics tools including mAbs, fusion proteins, and ADCs to target cancer and respiratory diseases. further i have worked on multicompartment & PBPK models to better understand PK parameters of antibody drugs in-vivo. 

 

Experience

Postdoctoral Associate, Pharmaceutical Sciences SUNY at Buffalo NY (July 2017 to May 2022)-

During my postdoctoral tenure, I worked in collaboration with Oncolinx, EVQLV, and Manhattan BioSolutions to develop novel Antibodies, Antibody-drug conjugates, scFv & FC-fusion proteins to target cancer, and mucosal antigens. 
1- Developed mAbs and antibody fragments, fusion proteins using CHO & EColi expression system.
2- Developed Antibody-drug conjugates using Oncolinx’s proprietary Azonafide molecules with cleavable & non-cleavable linker molecules. Further ADCs were developed using tubulin inhibitor (MMAE) & DNA intercalating toxins (PBDs). 
3- Developed analytical & bioanalytical methods to characterize and quantify ADCs, mAbs & attached and released payload in, Plasma and tissue matrix.
4- Analyzed NCA parametes using Winonlin & SimBiology to evaluate PK parameters of mAbs, and ADCs in animal models after IV administration.
5- developed Multi-compartment & PBPK models to perform model fitting and simulations of observed data from antibody & antibody fragments administered in mouse. 
6- Performed tumor regression experiments using xenograft and syngeneic mouse models in C57BL/6, Balb/C, and Nude mice.

Project Assistant Department of Pharmaceutics CSIR-CDRI Lucknow (Jan 2007 to March 2011)-


During my Project Assistant tenure, I developed nanoparticle-based formulations for improved delivery of proteins and small molecule drugs via oral, IV administration. The main focus of my research was to develop nanoparticle system that can protect the encapsulated protein from harsh environment outside of NPs with improved targeting & mucoadhesion to target tissues like cancer cells & mucosal as well as gut-associated lymph nodes. To achieve these goals, I have developed novel polymer conjugates using Lipids, Chitosan, and PLGA and conjugating them with cyanocobalamin & pyridoxine to improve the efficacy of the nanoparticle & liposome formulations. 
PK profile of developed formulations was evaluated by performing NCA using mouse and rat data. NCA parameters like AUC, CL, Volume & Bioavailability were calculated using WinNonlin & Simbiology.
